The Significance of P-glycoprotein, PCNA, DNA Content Cathepsin-D and Histological Grade as Prognostic Indicators of Brease Cancer.

Abstract
To evaluate the efficacy of P-glycoprotein, PCNA, DNA content and Cathepsin-D as prognostic indicators of breast cancer thirty cases of stage III breast cancer which were diagnosed and treated between 1986 were selected. Ten cases of benign
breast
disease were also studied for control. Breast cancer patients were allocate into recurrent group (15 cases) and non recurrent group (15 cases) within 3 years after surgery.
Immunohistochemical staining using labbelled streptavidin method was used for P-glycoprotein, PCNA and Cathepsin-D. P-glycoprotein was focally expressed in 8 out of 10 benign breast diseaes. The expression of P-glycoprotein was higher in cancer
than in
benign lesion. However, there was no statistically significant difference between recurrent and nonrecurren groups.
Paraffin embedded specimens were used for DNA content. All benign and nonrecurrent breast cancer specimens were diploid whereas 6 cases aneuploidy (40%) in recurrent group. Aneuploidy was more prevalent in patients who had more than 4 lymph nodes
metastasis S-phase and S+G2M fractions were significantly higher in breast cancer than in benign diseases. But there was no differnce between recurrent and nonrecurrent groups. The anaeuploidy in recurrent group was more frequently seen in
cancers
of
high histologic grade than in low grade.
The positivity of PCNA was higher in breast cancer than in benign lesion. In recurrent gorup. the positivity was higher than in nonrecurrent gorup.
There was no difference of Cathepsin-D positivity between recureent and nonrecurrent groups. As a result of this study, it is concluded that DNA ploidy, histologic grade, and PCNA considered to be useful prognostic factors in assessing the
recurrence.
However, P-glycoprotein, nCathepsin-D, and proliferative fractions are not useful.
